Definition
Peptide bioregulators are short peptides, largely developed at the St. Petersburg Institute of Bioregulation and Gerontology, proposed to increase an organism's resistance to extreme factors and to prevent accelerated aging and age-related diseases. [1] The concept originates from complex peptide preparations extracted from animal tissues such as the pineal preparation Epithalamin and the thymic preparation Thymalin, from which tissue-specific short peptides including the tetrapeptide Epitalon (Ala-Glu-Asp-Gly) were subsequently designed. [2] They underpin a proposed peptide theory of ageing, which frames ageing as an evolutionarily determined process of impaired synthesis of regulatory and tissue-specific peptides that can in principle be corrected by supplying such peptides. [2]
How it works
The proposed mechanism is that short peptides of 2 to 7 amino acid residues penetrate into cell nuclei and interact with the nucleosome, histone proteins, and single- and double-stranded DNA to regulate gene expression and protein synthesis. [3] Short peptides are proposed to regulate the status of DNA methylation, an epigenetic mechanism for activating or repressing genes in normal, pathological, and senescent states. [3] In preclinical work, peptides such as AEDG (epitalon) and KE (vilon) are reported to stimulate cell proliferation and differentiation, decrease apoptosis, and change expression of specific genes. Evidence & status
The evidence base is dominated by a single research tradition and consists largely of Russian preclinical and animal studies together with author-reported clinical trials, rather than independent large-scale randomized trials. [2] [1] In animal models, Epithalamin and its designed peptide Epitalon are reported to increase lifespan in rats, mice, and fruit flies and to restore circadian melatonin and cortisol rhythms in old rhesus monkeys. [2] Reviews from this group report that peptide injection increased the physiological resource of cells, tissues, and the organism by 20 to 42 percent and decreased mortality in elderly people, but these are review-level claims from the originating institute. [6]
Applications
Proposed applications include geroprotection and age-related disease prevention, with peptide preparations for the prostate, cerebral cortex, and eye retina reported to improve the condition of the respective organs. [2] Ultrashort peptides have been explored as candidate neuroprotective compounds in Alzheimer's disease via proposed epigenetic mechanisms, though this remains hypothesis-generating. [7] Some short peptides from this tradition have also been positioned as biologically active food additives (parapharmaceutics) for peroral administration. [1]
