Definition
Epithalon, also known as Epitalon or Epithalone, is a tetrapeptide, Ala-Glu-Asp-Gly (AEDG), synthesized based on the amino acid composition of Epithalamin, a bovine pineal gland extract. [1] Over the last 25 years it has been extensively studied using in vitro, in vivo, and in silico methods, with results indicating geroprotective and neuroendocrine effects. [1] It is classed among investigational, non-approved therapeutic peptides that show promising preclinical and limited clinical evidence but lack long-term safety data and systematic validation. [2]
How it works
Epithalon has been reported to exert a direct influence on melatonin synthesis, alter the mRNA levels of interleukin-2, modulate the mitogenic activity of murine thymocytes, and enhance the activity of various enzymes including telomerase, though it remains uncertain whether these are its sole mechanisms of action. [1] At the cellular level the AEDG peptide is described as regulating the expression of circadian-rhythm genes, reducing the synthesis of apoptosis-promoting proteins such as p16, p21 and p53, and regulating markers of functional cell activity including SIRT1 and SIRT6. [3] In animal studies Epitalon restored melatonin secretion in the pineal gland and had strong regulatory activity on the neuroimmunoendocrine and antioxidant systems of the organism. [4]
Evidence & status
In animal models Epitalon has been reported to increase the lifespan of mice and fruit flies and to restore the circadian rhythms of melatonin and cortisol production in old rhesus monkeys. [5] In geroprotection studies, the synthetic peptide bioregulator Epitalon was among substances that inhibited age-related alterations and spontaneous tumour development and showed life span extension in several mouse strains, with effects depending on dose, administration conditions, and genetic predisposition. [6] A 2026 gerontology review that identified epitalon under telomere biology concluded that non-approved investigational peptides showed promising preclinical and limited clinical evidence but require rigorous validation through well-designed clinical trials to establish safety and efficacy for healthspan extension. [2] Although preclinical studies of recovery-enhancing peptides such as epithalon are promising, there is a current lack of clinical trials. [7]
Considerations
Much of the evidence base for Epitalon derives from experimental and animal models, and it remains uncertain whether its reported mechanisms are the sole mechanisms of action. [1] As a non-FDA-approved investigational peptide, it lacks long-term safety data and systematic validation, and significant knowledge gaps include optimal dosing regimens, combination therapy effects, and biomarkers for monitoring efficacy. [2]
