Longevity Medicine
Definition
Longevity medicine brings fundamental ageing research together with clinical applications to advance the goal of promoting healthy longevity in the human population. [1] It is grounded in geroscience, a discipline that aims to define and modify aging-related biologic pathways, slow age-related disability, prevent age-related diseases, and increase disability-free survival. [2]
How it works
Its central premise, the geroscience hypothesis, proposes that addressing the biology of aging could directly prevent the onset or mitigate the severity of multiple chronic diseases. [3] Because age is the single most influential factor in many chronic conditions, targeting the ageing process itself is expected to have the greatest impact on human health. [4] The field is moving toward precision geromedicine, in which gerotherapeutics are anticipated to be tailored to each person based on their genetic profile, omics-based and clinical biomarkers of aging, psychosocial profile, and past or present exposures. [5]
Role in longevity
The proposed integration of generative AI, cutting-edge biological technology, and longevity medicine is put forward as essential for extending the productive and healthy human lifespan. [1] Biomarkers of aging are quantitative parameters that predict biological age and ideally its changes in response to interventions, giving the field the tools it needs to identify individuals aging faster and to evaluate longevity interventions. [6] [7]
Evidence & applications
Therapies that inhibit aging biology, such as caloric restriction, metformin, senolytics, or rapalogs, may slow the development and progression of disease and functional decline in humans. [2] In animal models caloric restriction increases mean lifespan from 10% to 40% compared with mice fed ad libitum, and in adults with obesity and diabetes randomization to caloric restriction was associated with a 15% reduction in all-cause mortality. [2] Clinical translation remains limited, in part due to the gap between preclinical research and the application of biomarkers of aging in clinical and other translational settings. [6]
