Definition
5-Amino-1MQ is a small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT), a metabolic enzyme that catalyzes the methylation of nicotinamide using the universal methyl donor S-adenosyl methionine, directly linking one-carbon metabolism with a cell's methylation balance and NAD+ levels. [1] NNMT is emerging as a key point of intersection between cellular metabolism and epigenetic gene regulation, and abnormal activation of the enzyme can disrupt the balance between energy and methylation potential, fueling diseases such as diabetes, obesity, fibrosis, and cancer. How it works
Small-molecule inhibitors targeting NNMT are designed to reduce elevated enzyme activity, and while NNMT exerts a beneficial effect by regulating lipid parameters in the liver, its expression in adipose tissue correlates with obesity and insulin resistance. [1] [2] NNMT represents an appealing druggable target for metabolic disorders as well as oncological and other diseases in which the protein is improperly activated. [1]
Evidence & status
The evidence for NNMT inhibitors is preclinical and experimental: efforts to develop them have historically been limited by insufficient target engagement, reduced bioavailability, and unknown safety profiles, with limited preclinical success across diseases. [2] More recent advances in NNMT small-molecule inhibitor development have yielded compounds with high affinities for cellular NNMT and promising safety profiles, but their therapeutic potential is still being assessed and current NNMT inhibitor drugs remain limited in clinical use.
